Gene targeting strategy for B-hTLR3 mice.
The exons 4~6 of mouse Tlr3 gene that encode the extracellular domain were replaced by human TLR3 exons 2~4 in B-hTLR3 mice.
Strain specific TLR3 expression analysis in homozygous B-hTLR3 mice by flow cytometry. Peritoneal washes and splenocytes were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hTLR3 mice (H/H). Flow cytometry analysis of the peritoneal washes-macrophages，splenocytes-macrophages，DCs were analyzed with anti-TLR3 antibody. Mouse TLR3 was detected in peritoneal washes-macrophages，Splenocytes-macrophages，DCs of wild-type mice. Human TLR3 was detected in peritoneal washes-macrophages，Splenocytes-macrophages，DCs of homozygous B-hTLR3 mice.
