Gene targeting strategy for B-hGFRAL mice. The exons 2-6 of mouse Gfral gene were replaced by human GFRAL exons 2-6 in B-hGFRAL mice.
Strain specific analysis of GFRAL gene expression in wild-type mice and B-hGFRAL mice by RT-PCR. Mouse Gfral mRNA was detectable in brain of wild-type C57BL/6 mice (+/+). Human GFRAL mRNA was detectable only in brain of homozygous B-hGFRAL mice but not in wild-type mice.
Strain specific analysis of GFRAL expression in C57BL/6 mice (+/+) and homozygous B-hGFRAL mice (H/H) by western blot. Ovary and stomach were collected from C57BL/6 mice (+/+) and homozygous B-hGFRAL mice (H/H). GFRAL was detectable in C57BL/6 mice (+/+) and B-hGFRAL mice (H/H) due to the cross-reactivity of antibodies.
