Gene targeting strategy for B-hCD95 mice.
The exons 2-7 of mouse Fas gene that encode the extracellular domain and transmembrane domain were replaced by human FAS exons 2-7 in B-hCD95 mice.
Strain specific CD95 expression analysis in homozygous B-hCD95 mice by flow cytometry. Splenocytes were collected from wild-type (WT) mice (+/+) and homozygous B-hCD95 mice (H/H), and analyzed by flow cytometry with species-specific anti-CD95 antibody. Mouse CD95 was detectable in WT mice (+/+). Human CD95 was exclusively detectable in homozygous B-hCD95 mice (H/H) but not in WT mice (+/+).
