Gene targeting strategy for B-hCD248 mice. The exon 1 of mouse Cd248 gene that encodes the extracellular region was replaced by human CD248 exon 1 in B-hCD248 mice.
Strain specific CD248 expression analysis in homozygous B-hCD248 mice by western blot.
Lung tissue was collected from wild-type C57BL/6 mice (+/+) and homozygous B-hCD248 mice (H/H), and analyzed by western blot with anti-CD248 antibody. Mouse CD248 was detectable in wild-type C57BL/6 mice and homozygous B-hCD248 mice, as the antibody has cross-reactivity with human CD248.
Strain specific analysis of CD248 gene expression in wild-type C57BL/6 mice and B-hCD248 mice by RT-PCR. Mouse Cd248 mRNA was detectable in the lung of wild-type mice (+/+). Human CD248 mRNA was detectable only in homozygous B-hCD248 mice (H/H) but not in wild-type mice.
