Protein expression analysis
Western blot analysis of ADORA2B protein expression in homozygous B-hADORA2B mice. Various tissue lysates were collected from wild-type C57BL/6 mice (+/+) and homozygous B-hADORA2B mice (H/H), and then analyzed by western blot with anti-ADORA2B antibody. 40 μg total proteins were loaded for western blotting analysis. hADORA2B was detected in heart, lung, kidney, brain, liver and small intestine from homozygous mice.
mRNA expression analysis
Strain specific analysis of ADORA2B mRNA expression in wild-type C57BL/6 mice and hADORA2B mice by RT-PCR. Brain RNA were isolated from wild type C57BL/6 mice (+/+) and homozygous B-hADORA2B mice (H/H). Mouse Adora2b mRNA was only detectable in wild-type C57BL/6 mice. Human ADORA2B mRNA was only detectable in homozygous B-hADORA2B mice but not in wild-type mice.
